Offer Description
Mucus hydrogel is the first innate defense covering secretory epithelia. This complex biological fluid is essential for the resident flora, trapping and eliminating pathogens and harmful particles. Gel-forming mucins are the main organic component of mucus. There are 5 of them, highly conserved between humans and mice, and encoded by the MUC2, 5AC, 5B, 6 and 19 genes. Numerous polymorphisms with potentially deleterious effects are found in these mucins. The group has demonstrated in vivo that MUC5B is a crucial mucin in the cervical canal to prevent vaginal pathogenic bacteria from gaining access to the uterine compartment, and thus prevent local inflammation which is the major cause of premature births.
The project aims to correlate loss of mucin function with the risk of preterm birth in mice, and to determine the abnormal properties of mucus using mouse experiments and cell culture (organ-on-a-chip). In addition to culture and classic molecular biology techniques, the techniques used include experimental infections, particle tracking and imaging.
