Main mission: 

Plant shoot branching is a developmental process which is strongly regulated by the environment. For example, nitrate availability has a promoting effect on shoot branching. However, the molecular mechanisms that underpin the regulation of shoot branching by nitrate remain elusive. NPF transporters play an important role in mediating the effect of nitrate in different development processes. Interestingly, the mutant npf4.4 displays an increased branching phenotype compared to the wild-type, especially under nitrate deficiency. The goal of this project is decipher how NPF4.4 is involved in the control of shoot branching. To achieve this, a fine characterisation at the genetic and molecular level will be performed. This will be combined with assays in a heterologous system (xenopus oocyte), aimed at testing the transport activity of this NPF for hormones, focusing on strigolactones, a class of hormones playing a crucial role in shoot branching.

The candidate will be asked to characterise the role of NPF4.4 in shoot branching and test the transport activity of selected NPFs for hormones involved in shoot branching, mainly focusing on strigolactones.

 

Activities: 

  • plant cultivation and phenotyping (shoot development), 
  • GUS staining, 
  • gene expression analysis (RNA-seq and Q-PCR), 
  • molecular cloning, 
  • work in xenopus oocyte (extrcating and injecting oocytes)
